I have a river on my layout, that’s painted and coated with acrylic gloss medium (4 layers). But now, it has a coat of dust on it. Can I just wipe it with a wet cloth? It’s fairly smooth.[?]
A soft cloth with some soapy water should work. So would a soft brush and a vacuum.

You might want to try Swiffer pads - they can be used dry and seem to have some feature that repels dust. I’ve been using them on an acrylic-covered coffee table, and on my TV and computer screens. I’ve seen a remarkable decrease in buildup, even on dust-magnet CRT screens.
Incidentally, that coffee table has artifacts in ‘valleys,’ sealed in by the acrylic; something like a model lake.
I use a soapy cloth after a quick vacuum.
